Abu Jahal cursed the Prophet ﷺ and his ancestors at length in public. When Hamzah, the Prophet ﷺ's uncle and a leading archer of Makkah, returned from an expedition, the women of Banu Hashim taunted him for not defending his nephew's honor. Enraged on learning it had happened publicly, Hamzah struck Abu Jahal across the face with his bow at the Ka'bah and declared himself a follower of the Prophet ﷺ's religion, though he had not planned to say so. After a night of anguished du'a, he went to the Prophet ﷺ the next morning, was convinced by him, and testified to the truth.
Hamzah was the most senior Qurayshi to convert up to that point, being the son of Abdul Muttalib and brother of Abu Talib; his conversion forced the mushrikeen to temper their open hostility toward the Muslims.
Umar accepted Islam three days after Hamzah, the answer to a prayer for one of two men.
The conversions of Hamzah and Umar, with the emigration, made the Quraysh feel threatened enough to cut off the Banu Hashim.

Who does he think he is to curse our men?
How dare you curse my own nephew?! And I too am a follower of his religion!
Leave him be, for I cursed his nephew like never before.
I testify you are speaking the Truth, and now I don't want to go back to the religion of my forefathers.
O Allah, you know I am one of the leaders of the Quraysh. And I have said something that I cannot take back. So if this matter is true, guide my heart to it. And if it is not true, cause me to die right now.
Said the night after Hamzah struck Abu Jahal and impulsively declared himself a follower of the Prophet's ﷺ religion, unsure whether he had spoken the truth.
